KAIST develops precise 3D brain-mimicking research platform

The Korea Advanced Institute of Science and Technology (KAIST) has developed a 3D brain-mimicking platform that achieves six times higher precision in replicating neuronal networks compared to conventional methods. According to KAIST, Professors Je-Kyun Park and Yoonkey Nam from the Department of Bio and Brain Engineering led the joint research effort to create a tool for studying brain structure and function simultaneously. 

The integrated platform uses low-viscosity natural hydrogels, which have mechanical properties similar to brain tissue, to create layered neuronal structures with high resolution. Traditional bioprinting relies on high-viscosity bioinks for structural stability but limits neuronal growth. In contrast, KAIST’s approach maintains biological compatibility while achieving precise patterning. 

Professor Je-Kyun Park stated, “This research is a joint development achievement of an integrated platform that can simultaneously reproduce the complex multilayered structure and function of brain tissue. Compared to existing technologies where signal measurement was impossible for more than 14 days, this platform maintains a stable microelectrode chip interface for over 27 days, allowing the real-time analysis of structure-function relationships.” 

The platform is based on three core technologies: 

• Capillary pinning effect: enables dilute hydrogels to adhere to a stainless-steel micromesh, producing brain-like structures with six times greater precision. 

• 3D printing aligner: ensures accurate stacking of printed layers, preventing misalignment and securing stable integration with microelectrode chips. 

• Dual-mode analysis system: measures electrical signals from below while simultaneously observing cell activity with calcium imaging from above. 

The research team successfully fabricated a three-layered mini-brain using fibrin hydrogel. Cortical neurons were placed in the upper and lower layers, with the middle layer left empty to allow neural connections to form. Electrical signals were recorded from the lower layer, while calcium imaging was performed on the upper layer. 

When electrical stimulation was applied, neurons in both layers responded simultaneously. After applying a synaptic blocker, the response decreased, confirming that the neurons were genuinely connected and transmitting signals. 

This technology could be used in various areas such as neurological disease modeling, brain function studies, neurotoxicity testing, and the development of neuroprotective drugs. Its ability to maintain stable signal measurement for over 27 days makes it a valuable tool for long-term brain research.
